Cagrilintide (development codes AM833 and NNC0174-0833) is a synthetic long-acting amylin analogue developed by Novo Nordisk, engineered from the amylin sequence with a fatty-diacid side chain that gives it a long, once-weekly-compatible half-life. It is also formulated with semaglutide as the investigational fixed-dose combination CagriSema.

A Phase 1b study (Enebo et al., 2021) characterised its pharmacokinetics alongside semaglutide, and a Phase 2 trial (Lau et al., 2021) examined once-weekly cagrilintide monotherapy in adults with overweight or obesity. The Phase 3 REDEFINE programme (Garvey et al., 2025) studied the CagriSema combination against each component alone.

Novo Nordisk submitted CagriSema to the FDA for weight management in December 2025; as of September 2026 neither cagrilintide alone nor CagriSema is an approved medicine in any jurisdiction. The FDA states that cagrilintide cannot be used in compounding under federal law and is not a component of any FDA-approved drug. Research-use-only vials sold commercially are outside Novo Nordisk’s clinical-trial supply chain and have not been reviewed for safety, effectiveness or quality.
